Yes, against the business account at the issuing bank. Some banks issue the consumer card on the business account instead; ask which product the account actually gets.

Is a fintech business account a substitute?

For separating spend, yes: Revolut Business and Qonto issue debit cards with controls. They are not credit; a credit line still needs a bank business card.
